چکیده

Corneal disease is a significant cause of decreased vision or blindness in the horse worldwide. Numerous topical and systemic drugs over a prolonged period of time may be used to treat an equine corneal ulcer. Despite the high prevalence of corneal disease and ulceration in the horse, and the complexity of corneal wound healing, research is limited and more heavily devoted to in vivo studies whose limitations include small sample sizes and ethical considerations. Bench top laboratory in vitro studies are limited in number and overlook the complexity of corneal wound healing by assessing each corneal layer separately from its neighbor. The objective of this study was to develop a novel ex vivo model of equine corneal wound healing that maintained structural integrity, and is physiologically relevant, mimicking in vivo events. In this randomized and controlled study, fourteen equine corneas were harvested within two hours of humane euthanasia for reasons unrelated to this study. All donors were screened and corneas were included if they were free of ocular and systemic disease. Corneas were immediately processed and corneoscleral rims excised 2 mm posterior to the limbus. Corneas were randomly assigned to one of the following groups: wounded (n=8) or unwounded (n=6) controls. Each pair of eyes was divided such that corneas from the same horse were assigned to a different group. In the event that only one of the two eyes was included, the eye was assigned to the wounded group. In the ‘wounded’ group, the axial cornea was wounded for 60 seconds

Effect of connexin 43 inhibition by the mimetic peptide Gap27 on corneal wound healing, inflammation and neovascularization

BACKGROUND AND PURPOSE The connexin 43 (Cx43) mimetic peptide Gap27 was designed to transiently block the function of this gap junction. This study was undertaken to investigate the effect of Gap27 on corneal healing, inflammation and neovascularization. EXPERIMENTAL APPROACH The effect of Gap27 on wound healing, inflammation and vascularization was assessed in primary human corneal epithelia...

ROCK Inhibitor Enhances Adhesion and Wound Healing of Human Corneal Endothelial Cells

Maintenance of corneal transparency is crucial for vision and depends mainly on the endothelium, a non-proliferative monolayer of cells covering the inner part of the cornea. When endothelial cell density falls below a critical threshold, the barrier and "pump" functions of the endothelium are compromised which results in corneal oedema and loss of visual acuity. The conventional treatment for ...

Nanofibrous Scaffolds for Orderly Corneal Wound Healing

Statement of Purpose: Corneal blindness deprives 10 million people worldwide of their sight. Current treatments for corneal blindness mainly rely on implants that replace the injured tissue and have been clinically unsuccessful thus far. The goal of this project is to prevent corneal blindness by developing clinically-viable scaffolds that promote orderly corneal wound healing using biochemical...

Effect of dexamethasone in combination with acetylcysteine at different times on corneal wound healing in dogs

The purpose of this study was to evaluate the effects of different time combinations of dexamethasoneand acetylcysteine on experimentally induced corneal ulcers in dogs. Experimental corneal wounds werecreated surgically to the anterior one third of the cornea in the center of all eyes of 15 mixed breed dogs. The eyes were divided into five groups according to planned post-operative medications...
